Indomethacin is a potent nonselective inhibitor of cyclooxygenase-1 and -2, sharply reducing prostaglandin synthesis. Lowering prostaglandin Eโ promotes ductus arteriosus constriction in preterm infants and provides robust anti-inflammatory and analgesic effects, but the potency also explains its higher rate of gastrointestinal and central nervous system adverse effects.
